β-caseínas A1 y A2 en leche bovina y bufalina: estado actual del conocimiento e implicaciones para la salud humana

Resumen

Antecedentes: La β-caseína es la segunda proteína más abundante de la leche de los mamíferos y presenta diversas variantes genéticas. Las isoformas A1 y A2 despiertan especial interés por sus implicaciones para la salud humana. Objetivo. Analizar críticamente la evidencia científica acerca de las variantes A1 y A2 de la β-caseína en la leche bovina y bufalina, sus posibles implicaciones para la salud humana y la distribución de sus genotipos en diferentes razas y especies. Desarrollo: La revisión narrativa incluyó artículos originales, revisiones sistemáticas y artículos de revisión que se localizaron en bases de datos con indización internacional. El análisis abarcó la caracterización molecular de la β-caseína, el origen evolutivo de las variantes A1 y A2, la formación de β-casomorfina-7, la evidencia experimental, clínica y epidemiológica acerca de sus posibles efectos en la salud humana y la distribución de los genotipos A1A1, A1A2 y A2A2 en razas bovinas y búfalos de agua. Conclusiones: La evidencia disponible confirma que la β-caseína A2 corresponde a la variante ancestral y que el búfalo de agua presenta predominantemente el genotipo A2A2. Aunque diversos estudios señalan posibles beneficios del consumo de leche A2, especialmente para la salud gastrointestinal, la relación entre la β-caseína A1, la β-casomorfina-7 y otras enfermedades crónicas permanece sin conclusión y requiere estudios clínicos con mayor rigor metodológico. La caracterización genética de los rebaños bovinos y bubalinos constituye una herramienta estratégica para los programas de mejoramiento genético, el desarrollo de productos lácteos con valor diferencial y la definición de prioridades de investigación.
